Probes and nano-delivery systems targeting NAD(P)H:quinone oxidoreductase 1: a mini-review

摘要: The two-electron cytoplasmic reductase NAD(P)H:quinone oxidoreductase 1 is expressed in many tissues. NAD(P)H:quinone oxidoreductase 1 is well-known for being highly expressed in most cancers. Therefore, it could be a target for cancer therapy. Because it is a quinone reductase, many bioimaging probes based on quinone structures target NAD(P)H:quinone oxidoreductase 1 to diagnose tumours. Its expression is higher in tumours than in normal tissues, and using target drugs such as β-lapachone to reduce side effects in normal tissues can help. However, the physicochemical properties of β-lapachone limit its application. The problem can be solved by using nanosystems to deliver β-lapachone. This mini-review summarizes quinone-based fluorescent, near-infrared and two-photon fluorescent probes, as well as nanosystems for delivering the NAD(P)H:quinone oxidoreductase 1-activating drug β-lapachone. This review provides valuable information for the future development of probes and nano-delivery systems that target NAD(P)H:quinone oxidoreductase 1.

Endemicity of H9N2 and H5N1 avian influenza viruses in poultry in China poses a serious threat to poultry

摘要: The H9N2 and H5N1 avian influenza viruses (AIVs) have been circulating in poultry in China and become endemic since 1998 and 2004, respectively. Currently, they are prevalent in poultry throughout China. This endemicity makes them actively involved in the emergence of the novel lineages of other subtypes of influenza viruses, such as the well-known viruses of the highly pathogenic avian influenza (HPAI) H5N2 and the 2013 novel H7N7, H7N9 and H10N8 subtypes, thereby threatening both the poultry industry and public health. Here, we will review briefly the prevalence and evolution, pathogenicity, transmission, and disease control of these two subtypes and also discuss the possibility of emergence of potentially virulent and highly transmissible AIVs to humans.

noncoding RNA LOC646029 functions as a ceRNA to suppress ovarian cancer progression through the miR-627-3p/SPRED1 axis

摘要: Long noncoding RNAs (lncRNAs) play a crucial regulatory role in the development and progression of multiple cancers. However, the potential mechanism by which lncRNAs affect the recurrence and metastasis of ovarian cancer remains unclear. In the current study, the lncRNA LOC646029 was markedly downregulated in metastatic ovarian tumors compared with primary tumors. Gain- and loss-of-function assays demonstrated that LOC646029 inhibits the proliferation, invasiveness, and metastasis of ovarian cancer cells in vivo and in vitro. Moreover, the downregulation of LOC646029 in metastatic ovarian tumors was strongly correlated with poor prognosis. Mechanistically, LOC646029 served as a miR-627-3p sponge to promote the expression of Sprouty-related EVH1 domain-containing protein 1, which is necessary for suppressing tumor metastasis and inhibiting KRAS signaling. Collectively, our results demonstrated that LOC646029 is involved in the progression and metastasis of ovarian cancer, which may be a potential prognostic biomarker.

Robust design approach to the minimization of functional performance variations of products and systems

摘要: Functional performance variations of products and systems are often used to measure the qualities of products and systems considering the changes in the design parameter values caused by uncertainties. A robust design approach has been developed in this research to minimize the functional performance variations considering the design parameter uncertainties by identifying the boundaries of the functional performance variations through optimization. In this work, a mathematical model is developed to describe the relationships among functional performance, design configurations and parameters, and design parameter uncertainties. A multi-level optimization model is established to identify: (1) The optimal design configuration, (2) the optimal values of design parameters, and (3) the boundaries of functional performance variations. Sensitivity analysis considering the impact of parameter uncertainties on functional performance variation boundaries has also been conducted. A case study on the design of a truss system has been conducted. Case study results show that the sensitivities of functional performance variation boundaries to the design parameter uncertainties can be reduced significantly using the new robust design approach.

Seasonal microbial community shift in a saline sewage treatment plant

摘要: Activated sludge was monthly sampled from a saline sewage treatment plant of Hong Kong (China) during June 2007 to May 2008 to analyze the microbial community shift along with environmental variations using denaturing gradient gel electrophoresis of polymerase chain reaction amplified 16S rDNA fragments. Environmental changes resulted into a seasonal microbial community shift characterized by alterations in species number and abundance in the sewage treatment plant. Correspondence analysis and cluster analysis on community structure profile showed that the 12 monthly samples fell into four groups, which is in accordance with season changing in Hong Kong. Canonical correspondence analysis revealed that and posed more significant effects on community structure than total phosphorus and total nitrogen, respectively. Compared with sludge retention time, influent total phosphorus had an inverse effect on the community structure shift, and chemical oxygen demand and showed a similar effects. Results of this study may contribute to the development of new knowledge involving the microbial community shift in sewage treatment plants.

The opportunity of membrane technology for hydrogen purification in the power to hydrogen (P2H) roadmap

摘要: The global energy market is in a transition towards low carbon fuel systems to ensure the sustainable development of our society and economy. This can be achieved by converting the surplus renewable energy into hydrogen gas. The injection of hydrogen (≤10% v/v) in the existing natural gas pipelines is demonstrated to have negligible effects on the pipelines and is a promising solution for hydrogen transportation and storage if the end-user purification technologies for hydrogen recovery from hydrogen enriched natural gas (HENG) are in place. In this review, promising membrane technologies for hydrogen separation is revisited and presented. Dense metallic membranes are highlighted with the ability of producing 99.9999999% (v/v) purity hydrogen product. However, high operating temperature (≥300 °C) incurs high energy penalty, thus, limits its application to hydrogen purification in the power to hydrogen roadmap. Polymeric membranes are a promising candidate for hydrogen separation with its commercial readiness. However, further investigation in the enhancement of H /CH selectivity is crucial to improve the separation performance. The potential impacts of impurities in HENG on membrane performance are also discussed. The research and development outlook are presented, highlighting the essence of upscaling the membrane separation processes and the integration of membrane technology with pressure swing adsorption technology.

NiCo2O4@quinone-rich N–C core–shell nanowires as composite electrode for electric double layer capacitor

摘要: The bind-free carbon cloth-supported electrodes hold the promises for high-performance electrochemical capacitors with high specific capacitance and good cyclic stability. Considering the close connection between their performance and the amount of carbon material loaded on the electrodes, in this work, NiCo2O4 nanowires were firstly grown on the substrate of active carbon cloth to provide the necessary surface area in the longitudinal direction. Then, the quinone-rich nitrogen-doped carbon shell structure was formed around NiCo2O4 nanowires, and the obtained composite was used as electrode for electric double layer capacitor. The results showed that the composite electrode displayed an area-specific capacitance of 1794 mF∙cm–2 at the current density of 1 mA∙cm–2. The assembled symmetric electric double layer capacitor achieved a high energy density of 6.55 mW∙h∙cm–3 at a power density of 180 mW∙cm–3. The assembled symmetric capacitor exhibited a capacitance retention of 88.96% after 10000 charge/discharge cycles at the current density of 20 mA∙cm–2. These results indicated the potentials in the preparation of the carbon electrode materials with high energy density and good cycling stability.

A pre-compensation method of the systematic contouring error for repetitive command paths

摘要:

For a repetitive command path, pre-compensating the contouring error by modifying the command path is practical. To obtain the pre-compensation value with better accuracy, this paper proposes the use of a back propagation neural network to extract the function of systematic contouring errors. Furthermore, by using the extracted function, the contouring error can be easily pre-compensated. The experiment results verify that the proposed compensation method can effectively reduce contouring errors.

Synthesis of zeolite Na-P1 from coal fly ash produced by gasification and its application as adsorbent

摘要: The coal fly ash produced by gasification is estimated to be over 80 million ton per year in China by 2021. It has mainly been disposed as solid waste by landfill. There is lack of study focused on its utilization. In this paper, the coal fly ash produced by gasification was at first analyzed and then applied to synthesize zeolite as an adsorbent. The effects of synthesis conditions on the cation exchange capacity (CEC) of zeolite were investigated. The results from X-ray diffraction and scanning electron microscope indicated that the crystallinity of the synthesized zeolite is the most important factor to affect the CEC. When the synthesized zeolite with the highest CEC (275.5 meq/100 g) was used for the adsorption of Cr(VI) from aqueous solution, the maximum adsorption capacity for Cr(VI) was found to be 17.924 mg/g. The effects of pH, contact time and initial concentration on the adsorption of Cr(VI) were also investigated. The adsorption kinetics and isotherms can be well described by the pseudo-second-order model and Langmuir isotherm model, respectively.

摘要: A series of new benzimidazole derivatives were designed and synthesized. Their chemical structures were testified by H NMR, infrared spectroscopy (IR), mass spectrography (MS), and elemental analysis. Their potent antiviral properties indicated the prospect of new drugs. Compound , , , , , , and were identified as novel antivirus with much better selective activity and inhibitory activity than the comparable ribavirin against Coxsackie virus B in VERO cells.

We review here the novel cardiac protective effects of the multifunctional enzyme, p21-activated kinase 1 (PAK1), a member of a serine/threonine protein kinase family. Despite the large body of evidence from studies in noncardiac tissue indicating that PAK1 activity is key in the regulation of a number of cellular functions, the role of PAK1 in the heart has only been revealed over the past few years. In this review, we assemble an overview of the recent findings on PAK1 signaling in the heart, particularly its cardiac protective effects. We present a model for PAK1 signaling that provides a mechanism for specifically affecting cardiac cellular processes in which regulation of protein phosphorylation states by protein phosphatase 2A (PP2A) predominates. We discuss the anti-adrenergic and antihypertrophic cardiac protective effects of PAK1, as well as its role in maintaining ventricular Ca2+ homeostasis and electrophysiological stability under physiological, β-adrenergic and hypertrophic stress conditions.

A universal vaccine is in high demand to address the uncertainties of antigenic drift and the reduced effectiveness of current influenza vaccines. In this study, a strategy called computationally optimized broadly reactive antigen (COBRA) was used to generate a consensus sequence of the hemagglutinin globular head portion (HA1) of influenza virus samples collected from 1918 to 2021 to trace evolutionary changes and incorporate them into the designed constructs. Constructs carrying different HA1 regions were delivered into eukaryotic cells by Salmonella-mediated bactofection using a Semliki Forest virus RdRp-dependent eukaryotic expression system, pJHL204. Recombinant protein expression was confirmed by Western blot and immunofluorescence assays. Mice immunized with the designed constructs produced a humoral response, with a significant increase in immunoglobulin G (IgG) levels, and a cell-mediated immune response, including a 1.5-fold increase in CD4+ and CD8+ T cells. Specifically, constructs #1 and #5 increased the production of interferon-γ (IFN-γ) producing CD4+ and CD8+ T cells, skewing the response toward the T helper type 1 cell (Th1) pathway. Additionally, interleukin-4 (IL-4)-producing T cells were upregulated 4-fold. Protective efficacy was demonstrated, with up to 4-fold higher production of neutralizing antibodies and a hemagglutination inhibition titer > 40 against the selected viral strains. The designed constructs conferred a broadly protective immune response, resulting in a notable reduction in viral titer and minimal inflammation in the lungs of mice challenged with the influenza A/PR8/34, A/Brisbane/59/2007, A/California/07/2009, KBPV VR-92, and NCCP 43021 strains. This discovery revolutionizes influenza vaccine design and delivery; Salmonella-mediated COBRA-HA1 is a highly effective in vivo antigen presentation strategy. This approach can effectively combat seasonal H1N1 influenza strains and potential pandemic outbreaks.
